Proteintech offers immunization of two rabbits for customers who have already prepared antigens (Catalog# 90001).  The customer can choose from one of the following standard protocols for immunization. We recommend customers opt for the long protocol as it has a higher success rate - the short protocol is designed for the needs of customers who need an antibody quickly and are willing to trade success rate for time.
Guarantee: Customers will only be charged when ELISA or Dot assay show a positive response to the antigen.

- Which Protocol should I use?
The standard long protocol is the preferred protocol because it has been widely used and tested through history. The long protocol provides a better success rate than the short protocol. The short protocol is designed for customers who need the antibody in a rush and are willing to trade success rate for time.
- How much antigen do you need?
For a protein antigen, the optimum amount is 1 mg of protein for each rabbit (2 mg for two rabbits for each order). The minimum amount is 300 ug/per rabbit. For a peptide antigen, a minimum of 5 mg of peptide conjugated to 2.5 mg of KLH/per rabbit will be needed for immunization (In Proteintech's peptide antibody protocol, we use 10 mg of peptide to conjugate 5 mg of KLH).
- In what form should I send the protein to Proteintech?
The antigen can be in any biological buffer without detergent or Urea. It can be sent frozen or at 4°C by customer's discretion.
- Can I send you proteins in gel slices cut off from SDS PAGE?
Yes, protein in gel slices can also be used as an antigen, to a lesser extent as a soluble or precipitated antigen.
- How much antiserum will I receive?
Each rabbit will give ~12 ml of serum in each production bleed and 45-50 ml of serum from 90 - 100 ml of blood in the final bleed, totaling about 150 ml of antiserum from two rabbits.
- I do not have an antigen; can Proteintech synthesize a peptide or make the fusion protein?

- Can precipitated protein be used as an antigen?
Yes. In many cases we have observed that a precipitated protein yields a better antibody than a soluble antigen. The speculation is that the precipitated antigens provide a long term antigen depot for continuing stimulation of the immune system which may also be easier for an antigen presenting cells to engulf.
