Service Description:

Proteintech offers immunization of two rabbits and affinity purification for those customers who have already prepared antigens. The customer can choose from one of two protocols for immunization. Proteintech will prepare an affinity purification column and purify 30 ml of antiserum with the column. The customer will receive the column, purified anti-serum, flow through and the rest of the anti serum.

FAQs:

- Which protocol should I use?
The standard protocol is the preferred protocol because it has been widely used and tested thoroughly. The long protocol provides a better success rate than the short protocol. The short protocol is designed for customers who need the antibody in a rush and are willing to trade success rate for time.
- How much antigen do you need?
For a protein antigen, 3 mg of protein will be needed for each column. For a peptide antigen, 15 mg will be needed.
- In what form should i send the protein to Proteintech?
The antigen used for immunization can be in any biological buffer without detergent or Urea, it can also be precipitated or gel slices from an SDS PAGE. It is also requested that you also provide a soluble antigen for purification in a non-NH2 buffer.
- How much antiserum will I receive?
Each rabbit will give approximately 12ml of serum in each production bleed and 45 - 50 ml of serum from 90 - 100 ml of blood in the final bleed, therefore on average a total of 150ml is produced from 2 rabbits. 30 ml of this will be used for purification and the rest of the serum is sent to you.
- I do not have an antigen; can Proteintech synthesize a peptide or make the fusion protein?

- How does Proteintech determine which rabbit serum to purify?
It is decided by the ELISA result from the test bleed. If both rabbits give both results, we purify 15 ml of serum from each rabbit. If one rabbit gives better results than the other then we purify 30 ml from the 'good' rabbit.
- How much purified antibody will I receive?
It depends on the concentration of the specific antibody in the serum provided. Usually we can get 2-10 ml from 15 ml of antiserum.
