Product Information

KE10062 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The MFG-E8 ELISA kit is to be used to detect and quantify protein levels of endogenous MFG-E8. The assay recognizes mouse MFG-E8. An antibody specific for MFG-E8 has been pre-coated onto the microwells. The MFG-E8 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse MFG-E8 is added to detect the captured mouse MFG-E8 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

Milk fat globule-epidermal growth factor-factor 8 (MFG-E8), also known as lactadherin, is a multifunctional secreted glycoprotein exhibiting versatile functions in cell physiology affecting health and diseases. It plays roles in clearance of apoptotic cells, anti-inflammatory, wound healing, arterial remodeling, angiogenesis, enhancing tumorigenicity and cancer metastasis. MFG-E8 consists of two-repeated EGF-likedomains, a mucin-like domain and two-repeated discoidin-like domains (C-domains), and contains an integrin-binding motif (RGD sequence) in the EGF-like domain. It shows ubiquitous pattern of expression in various types of cells and tissues. Altered expression of MFG-E8 causes disruptions of homeostasis and is correlated with a number of diseases.
