Background Information
ANKZF1, the mammalian ortholog of yeast Vms1, functions in the ribosome quality control (RQC) pathway as a tRNA hydrolase that recycles nascent peptides from stalled ribosomes, and it also counterbalances Rqc2‑mediated CAT‑tailing, thereby limiting the import of CAT‑tailed mitochondrial precursor proteins and reducing their aggregation within mitochondria. Under oxidative stress, both Vms1 and ANKZF1 localize to mitochondria and recruit Cdc48/VCP to facilitate mitochondria‑associated degradation (MAD), although the physiological requirement for this mitochondrial targeting remains unclear. Notably, recent studies in yeast reveal that VMS1 deletion aggravates proteotoxicity in the mitochondrial matrix, highlighting its essential protective role during mitochondrial stress.
