Background Information

MBT domain-containing protein 1 (MBTD1) is a chromatin-associated protein characterized by an N-terminal FCS-type zinc finger and four Malignant Brain Tumor (MBT) repeats at its C-terminus (PMID: 19841675). The MBT domains enable selective binding to mono- and dimethylated histone H4 lysine 20 (H4K20me1/2), functioning as a transcriptional repressor (PMID: 37523546). MBTD1 is a core subunit of the NuA4/TIP60 histone acetyltransferase complex, which plays a critical role in chromatin remodeling and transcriptional activation (PMID: 35705031). Aberrant chromosomal rearrangements involving MBTD1, such as the ZMYND11-MBTD1 fusion in acute myeloid leukemia and the MBTD1-CXorf67 fusion in endometrial stromal sarcoma, highlight its emerging significance in oncogenesis (PMID: 33594072, PMID: 23959973).
