Background Information
Platelet-activating factor acetylhydrolase 2 (PAFAH2), also known as PAF-AH II, is an intracellular phospholipase A2 enzyme that hydrolyzes platelet-activating factor (PAF) and oxidatively fragmented phospholipids (PMID: 34116047). Belonging to the PAFAH family, PAFAH2 shares significant homology with plasma PAFAH (PLA2G7) but is distinct in its intracellular localization (PMID: 8955149). The enzyme features an α/β-hydrolase fold with a Ser-His-Asp catalytic triad and does not require Ca²⁺ for activity (PMID: 34116047). PAF-AH II expresses in almost all tissues, but predominantly in epithelial cells such as renal proximal and distal tubules, hepatocytes, and intestinal column epithelium. PAFAH2 isolated from mammalian cells was found to be both a monomer and dimer. In an unstressed situation, PAFAH2 as a dimer remains in the cytoplasm, and when exposed to oxidative stress, the homodimers of PAFAH2 dissociate into monomers to bind to membranes, ER, and Golgi apparatus (PMID: 34116047).
