Background Information
PATZ1 (POZ/BTB and AT-hook containing Zinc finger protein 1) is an important multifunctional transcriptional regulator. Its protein structure is distinctive, containing a POZ/BTB domain (which mediates protein-protein interactions and transcriptional repression) and an AT-hook motif (which binds to AT-rich DNA sequences), as well as multiple C2H2-type zinc finger domains (responsible for sequence-specific DNA binding). PATZ1 plays a "double-edged sword" role in regulating gene expression, capable of either activating or repressing target gene transcription depending on the co-regulators it interacts with and the cellular context. It plays a critical role in embryonic development and in the maintenance of stem cell pluripotency and differentiation. In recent years, its dual role in cancer has garnered significant attention. In various tumors (such as glioma, sarcoma, and colorectal cancer), PATZ1 can function as either an oncogene or a tumor suppressor depending on the context, participating in tumorigenesis and progression by influencing pathways such as the cell cycle, apoptosis, and the DNA damage response.
