Background Information
Peroxisome assembly protein 12 (PEX12), also known as Peroxisome assembly factor 3 (PAF-3), is an integral peroxisomal membrane protein that is essential for peroxisomal matrix protein import and is one of only two peroxins known to interact with the ligand-binding domain of PEX5. Mutations in human PEX12 result in Zellweger syndrome, a lethal neurological disorder, and implicate the zinc ring domain in PEX12 function (PMID: 12456682). Pex12 exhibits protein-Ub ligase activity with specificity for the E2 protein Pex4 and Ubc4 (PMID: 19687296).
