Product Information

KE10068 is a solid phase sandwich Enzyme Linked-Immuno-Sorbent Assay (Sandwich ELISA). The mouse IL-12/IL-23 p40 ELISA kit is to be used to detect and quantify protein levels of endogenous mouse IL-12/IL-23 p40. The assay recognizes mouse IL-12/IL-23 p40. An antibody specific for mouse IL-12/IL-23 p40 has been pre-coated onto the microwells. The mouse IL-12/IL-23 p40 protein in samples is captured by the coated antibody after incubation. Following extensive washing, another antibody of biotinylated specific for mouse IL-12/IL-23 p40 is added to detect the captured mouse IL-12/IL-23 p40 protein. For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent. Solution containing sulfuric acid is used to stop color development and the color intensity which is proportional to the quantity of bound protein is measurable at 450 nm with the correction wavelength set at 630 nm.

Background Information

IL-12p40 is known as a component of the bioactive cytokines interleukin IL-12 and IL-23, which can exist in a monomeric or homodimeric form. IL-12p40 is produced primarily by activated inflammatory cells including macrophages, neutrophils, microglia and dendritic cells (DCs). The most widely appreciated function of IL-12p40 is to provide a negative feedback loop by competitively binding to the IL-12 receptor. Additionally, IL-12p40 can also act as a chemoattractant for macrophages and promote the migration of bacterially stimulated dendritic cells. It is also associated with several pathogenic inflammatory responses such as silicosis, graft rejection.
