Magnetic Cell Separation Systems

Magnetic Cell Separation Systems

The characterisation of specific cell types and their use in various downstream applications requires that cells of interest can be isolated from a heterogeneous cell population such as PBMC or whole blood. Proteintech have developed magnetic cell separation systems using our highly specific, high-affinity antibodies (or streptavidin) covalently conjugated to supramagnetic polystyrene particles.

Every kit has been completely developed, manufactured, and fully-validated in-house from the ground up to facilitate fast, straightforward cell separations.

Simply incubate your heterogeneous cell suspension with Proteintech’s magnetic cell separation particles to discover quick, easy, column-free cell depletions and isolations, resulting in a high purity homogeneous cell population!

Proteintech’s antibody quality and in-house validation you know and trust.

High lot-to-lot consistency.

Routinely tested in peripheral blood mononuclear cells and whole blood.

User-friendly, fast and easy to use compared to cell separation by fluorescence activated cell sorting (FACS).

Tested with multiple magnets; you can use the magnet you already have!

A chosen cell type can be isolated/depleted from a heterogeneous cell suspension through targeting with a specific antibody-magnetic particle complex directed at a target antigen on the desired cell type. This system uses Proteintech’s high specificity antibodies bound to supramagnetic particles (or biotin-streptavidin supramagnetic particle complex). This complex can target and bind to the desired cell surface antigen with high affinity, so that when placed inside a separation magnet the bound cells are attracted to the side of the tube, leaving the unbound cells to be transferred to a different tube.

downstream beads data cd4

Enrichment of CD4+ T lymphocytes using Human CD4 Magnetic Beads (Proteintech, MS003): CD4+ T cells isolated by magnetic selection from healthy donor PBMC were labeled with CellTrace violet (Thermofisher) to track cell division and stimulated in vitro for 72 hours with plate-bound anti-CD3 (1ug/mL, Proteintech, 65133-1-Ig) and CD28 (2ug/mL, Proteintech 65099-1-Ig). CD4+ T lymphocytes show high purity and active cell division after in vitro stimulation.

Q: Can I do flow cytometry after using the beads?

A: You can use the ‘untouched’ cells in flow cytometry, but not the cells bound to beads. You can use either the touched or the untouched in in vitro cell culture and downstream assays.

Q: Are the beads internalized by the cells?

A: No, the beads are not internalized by the cells.

Q: Do you have untouched cell isolation kits?

A: They will be released in 2022.

Q: Which magnet can I use your products with?

A: We have tested these products with a range of magnets, you can use the one you currently have in your lab!

Q: Do Proteintech have a magnet for cell separations?

A: This is currently in discussion/development.

Q: What is the size of the magnetic particles?

A: The diameter is approximately 2.7µM.

Q: Can I use these products to isolate cells from whole blood?

A: Yes, we have tested these products in whole blood, please follow the QR code on your protocol to download the whole blood protocol.

Q: Why would I use these instead of FACS or column-based MACS?

A: The protocol for Proteintech Magnetic Beads and Magnetic Beads Kits faster, and simpler than either FACS or column-based MACS, with fewer washing steps involved. The cells have undergone a gentler protocol during isolations.

Q: What is the shelf-life of the product?

A: The shelf life is 12 months at 4℃.

Q: Can I remove the magnetic beads from the bound cells?

A: We do not currently have magnetic beads with a release complex, this is currently in development.

Q: What is the recommended separation buffer?

A: PBS, 0.1% BSA, 2mM EDTA, pH 7.4 - you can also find this on your protocol.

High Purity Cell Depletions

Following depletion of CD8+ cells, supernatant cell suspension was stained with PB450-CD3(clone: HIT3a) and CL647-CD8(clone: OKT8) antibodies. CD45 positive cells are gated in the analysis. Left panel: CD3+CD8+ cells before selection. Right panel: CD3+CD8+ cells after depletion. Human CD8 magnetic beads are tested using PBMC from three different donors.
