  Various lysates were subjected to SDS PAGE followed by western blot with 84417-5-RR (CD157 antibody) at dilution of 1:4000 incubated at room temperature for 1.5 hours.

  Immunohistochemical analysis of paraffin-embedded mouse spleen tissue slide using 84417-5-RR (CD157 antibody) at dilution of 1:200 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  Immunofluorescent analysis of (4% PFA) fixed RAW 264.7 cells using CD157 antibody (84417-5-RR, Clone: 241822F10 ) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2).

  Affinity and Kinetic Characterization of 84417-5-RR

  Biolayer interferometry (BLl) kinetic assays of 84417-5-RR against Mouse CD157 were performed. The affinity constant is 5.36 nM.

CD157 is a glycosyl phosphotidylinositol-anchored protein that stimulates pre-B-cell growth and has adenosine diphosphate (ADP)-ribosyl cyclase and cyclic ADP-ribose (cADPR) hydrolase activity (PMID: 9473467, 11866528). The two enzymatic activities are responsible for the synthesis and hydrolysis of cADPR, a novel second messenger of calcium release from intracellular calcium stores. CD157 has a calculated molecular mass of 30-36 kDa and an apparent molecular mass of 40-46 kDa due to glycosylation (PMID: 8941363, 10491089).
