  Various lysates were subjected to SDS PAGE followed by western blot with 84139-4-RR (CD3EAP antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  FC experiment of U2OS using 84139-4-RR

  1x10^6 U2OS cells were intracellularly stained with 0.25 ug CD3EAP Recombinant antibody (84139-4-RR, Clone:241379H5) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set.

  Affinity and Kinetic Characterization of 84139-4-RR

  Biolayer interferometry (BLl) kinetic assays of 84139-4-RR against Human CD3EAP were performed. The affinity constant is 0.339 nM.

Human RNA polymerase I (Pol I)-specific subunit, previously identified as ASE-1 and as CD3ɛ-associated signal transducer (CAST), CD3EAP is a 55 kDa nucleolar autoantigen that has an apparent molecular mass of 90 kDa (PMID: 11199923). CD3EAP/hPAF49 contains a single tyrosine residue at position 82 (Tyr82), which is phosphorylated upon stimulation of the T-cell receptor. Western blot analysis detected CD3EAP at an apparent molecular mass of ~80-90 kDa.
