  WB analysis using 85013-4-RR (same clone as 85013-4-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 85013-4-RR (CPT2 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 85013-4-PBS in a different storage buffer formulation.

  IHC staining of mouse liver using 85013-4-RR (same clone as 85013-4-PBS)

  Immunohistochemical analysis of paraffin-embedded mouse liver tissue slide using 85013-4-RR (CPT2 antibody) at dilution of 1:500 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 85013-4-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 85013-4-RR

  Biolayer interferometry (BLl) kinetic assays of 85013-4-RR against Human CPT2 were performed. The affinity constant is 0.261 nM.

Hintergrundinformationen

Carnitine palmitoyltransferase-1 and -2 (CPT1 and CPT2) are two genetically distinct mitochondrial membrane bound enzymes and play critical role in the regulation of FAO in normal cells. CPT2 is an ubiquitous protein and locates in the inner membrane of mitochondrial (PMID: 29437870). CPT2 is frequently down-regulated in primary ovarian serous carcinomas, which is significantly correlated with poor survival of ovarian cancer patients (PMID: 33486313). Down-regulation of CPT2 was a major cause of acylcarnitine accumulation and a common feature in mouse models of obesity- and NASH-driven HCC and human SH-HCC (PMID: 29872321).
