  WB analysis using 67915-1-Ig

  Various lysates were subjected to SDS PAGE followed by western blot with 67915-1-Ig (DDX3 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated GAPDH Monoclonal antibody (HRP-60004) as loading control.

  WB result of DDX3 antibody (67915-1-Ig; 1:2000; incubated at room temperature for 1.5 hours) with sh-Control and sh-DDX3 transfected HepG2 cells.

  Immunohistochemical analysis of paraffin-embedded human breast cancer tissue slide using 67915-1-Ig (DDX3 antibody) at dilution of 1:500 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  FC experiment of HepG2 using 67915-1-Ig

  1X10^6 HepG2 cells were intracellularly stained with 0.5 ug Anti-Human DDX3 (67915-1-Ig, Clone:1D3A1) and CoraLite®488-Conjugated AffiniPure Goat Anti-Mouse IgG(H+L) at dilution 1:1000 (red), or 0.5 ug Mouse IgG2b Isotype Control (66360-3-Ig, Clone: K11B8C4B5) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

Geprüfte Anwendungen

• Erfolgreiche Detektion in WB: LNCaP-Zellen, HEK-293-Zellen, HeLa-Zellen, HepG2-Zellen, Jurkat-Zellen
• Erfolgreiche Detektion in IHC: humanes Mammakarzinomgewebe; Hinweis: Antigendemaskierung mit TE-Puffer pH 9,0 empfohlen. (*) Wahlweise kann die Antigendemaskierung auch mit Citratpuffer pH 6,0 erfolgen.
• Erfolgreiche Detektion in FC (Intra): HepG2-Zellen

Hintergrundinformationen

DDX3X, also named DBX, DDX3, and HLP2, belongs to the DEAD-box helicase family and DDX3/DED1 subfamily. It is ATP-dependent RNA helicase. DDX3X acts as a cofactor for XPO1-mediated nuclear export of incompletely spliced HIV-1 Rev RNAs. It is also involved in HIV-1 replication. DDX3X interacts specifically with hepatitis C virus core protein resulting in a change in intracellular location. 73kd is the target band. 60-65kd is some other isoform. This antibody can bind both DDX3X and DDX3Y for the close sequences.
