  WB analysis of U-87 MG using 84018-4-RR (same clone as 84018-4-PBS)

  U-87 MG cells were subjected to SDS PAGE followed by western blot with 84018-4-RR (FAP antibody) at dilution of 1:4000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 84018-4-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed Transfected HEK-293 cells using FAP antibody (84018-4-RR, Clone: 241129B4 ) at dilution of 1:400 and CoraLite®594-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-4). This data was developed using the same antibody clone with 84018-4-PBS in a different storage buffer formulation.

  FC experiment of Transfected HEK-293T using 84018-4-RR (same clone as 84018-4-PBS)

  1x10^6 HEK-293T cells or FAP-transfected HEK-293T cells were surface stained with 0.25 ug FAP Recombinant antibody (84018-4-RR, Clone: 241129B4) and Multi-rAb CoraLite ® Plus 647-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L) (RGAR005). Cells were not fixed. This data was developed using the same antibody clone with 84018-4-PBS in a different storage buffer formulation.

  1x10^6 untransfected (left) or FAP transfected (right) HEK-293T cells were surface stained with 0.25 ug FAP Recombinant antibody (84018-4-RR, Clone:241129B4) (red) or Isotype Control (blue), and Multi-rAb CoraLite ® Plus 647-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L) (RGAR005). 1x10^6 HEK-293T cells were surface stained with 0.25 ug FAP Recombinant antibody (84018-4-RR, Clone:241129B4) (red), or Isotype Control (blue) and Multi-rAb CoraLite ® Plus 647-Goat Anti-Rabbit Recombinant Secondary Antibody (H+L) (RGAR005).   Affinity and Kinetic Characterization of 84018-4-RR

  Biolayer interferometry (BLl) kinetic assays of 84018-4-RR against Human FAP were performed. The affinity constant is 0.126 nM.

Hintergrundinformationen

Fibroblast activation protein-alpha (FAP-alpha) is a cell surface glycoprotein serine protease, which can participate in extracellular matrix degradation and is involved in many cellular processes including tissue remodeling, fibrosis, wound healing, inflammation and tumor growth (PMID: 14751930; 16223769). FAP-alpha is a single transmembrane protein that locates the cytoplasm and plasma membrane. Immunohistochemical studies have shown that FAP-alpha is transiently expressed in certain normal fetal mesenchymal tissues but that normal adult tissues as well as malignant epithelial, neural, and hematopoietic cells are generally FAP-alpha-negative. However, most of the common types of epithelial cancers contain abundant FAP-alpha-reactive stromal fibroblasts (PMID: 7911242).
