  IHC staining of mouse skeletal muscle using 82941-1-RR (same clone as 82941-1-PBS)

  Immunohistochemical analysis of paraffin-embedded mouse skeletal muscle tissue slide using 82941-1-RR (FBXO32 antibody) at dilution of 1:200 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 82941-1-PBS in a different storage buffer formulation.

  IF Staining of C2C12 using 82941-1-RR (same clone as 82941-1-PBS)

  Immunofluorescent analysis of (4% PFA) fixed C2C12 cells using FBXO32 antibody (82941-1-RR, Clone: 230243H7 ) at dilution of 1:250 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 82941-1-PBS in a different storage buffer formulation.

  FC experiment of U-251 using 82941-1-RR (same clone as 82941-1-PBS)

  1x10^6 U-251 cells were intracellularly stained with 0.25 ug Anti-Human FBXO32 (82941-1-RR, Clone:230243H7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug rabbit IgG isotype control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set. This data was developed using the same antibody clone with 82941-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 82941-1-RR

  Biolayer interferometry (BLl) kinetic assays of 82941-1-RR against Human FBXO32 were performed. The affinity constant is below 1 pM.

Hintergrundinformationen

FBXO32 (F box only protein 32), also known as Atrogin 1 or MAFbx, is a member of the F-box protein family which is characterized by an approximately 40 amino acid motif F-box. This protein is an E3 ubiquitin ligase that is markedly up-regulated in muscle atrophy. FBXO32 is thus a potential drug target for the treatment of muscle atrophy. Some data support that FBXO32 may play an important role in tumorigenesis. Recent study reveal that FBXO32 targets the oncogenic protein c-Myc for ubiquitination and degradation through the proteasome pathway.
