  WB analysis of HeLa using 83509-6-RR (same clone as 83509-6-PBS)

  WB result of FBXO38 antibody (83509-6-RR; 1:1000; incubated at room temperature for 1.5 hours) with sh-Control and sh-FBXO38 transfected HeLa cells. This data was developed using the same antibody clone with 83509-6-PBS in a different storage buffer formulation.

  Various lysates were subjected to SDS PAGE followed by western blot with 83509-6-RR (FBXO38 antibody) at dilution of 1:3000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83509-6-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 83509-6-RR (same clone as 83509-6-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug FBXO38 Recombinant antibody (83509-6-RR, Clone:240513C9) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83509-6-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83509-6-RR

  Biolayer interferometry (BLl) kinetic assays of 83509-6-RR against Human FBXO38 were performed. The affinity constant is 0.723 nM.

FBXO38, also known as HMN2D, MOKA, and SP329, is a protein that plays a significant role in the regulation of immune responses, particularly in the context of cancer and autoimmune diseases. FBXO38 has been identified as an E3 ubiquitin ligase that mediates the degradation of PD-1 (Programmed cell death protein 1), a key immune checkpoint protein expressed on the surface of T cells. The internalization, ubiquitination, and proteasome degradation of PD-1 are facilitated by FBXO38, which links PD-1 to Lys48, leading to its poly-ubiquitination and subsequent degradation (PMID: 30487606).
