  WB analysis using 83624-1-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 83624-1-RR (HDAC1 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  IHC staining of mouse colon using 83624-1-RR

  Immunohistochemical analysis of paraffin-embedded mouse colon tissue slide using 83624-1-RR (HDAC1 antibody) at dilution of 1:1000 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  Immunofluorescent analysis of (4% PFA) fixed HeLa cells using HDAC1 antibody (83624-1-RR, Clone: 240614A3 ) at dilution of 1:250 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red).

  FC experiment of HeLa using 83624-1-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug HDAC1 Recombinant antibody (83624-1-RR, Clone:240614A3) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

  ChIP experiment of K-562 using 83624-1-RR

  Chromatin was prepared from K-562 cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of HDAC1 (83624-1-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR.

  Affinity and Kinetic Characterization of 83624-1-RR

  Biolayer interferometry (BLl) kinetic assays of 83624-1-RR against Human HDAC1 were performed. The affinity constant is below 1 pM.

Histone deacetylases(HDAC) are a class of enzymes that remove the acetyl groups from the lysine residues leading to the formation of a condensed and transcriptionally silenced chromatin. The protein encoded by this gene belongs to the histone deacetylase/acuc/apha family and is a component of the histone deacetylase complex, which is responsible for gene expression silencing. It also plays an important role in the control of cell proliferation and differentiation by interacting with RB, p53 and other transcription factors. At least 4 classes of HDAC were identified. As a class I HDAC, HDAC 1 was primarily found in the nucleus.
