  Various lysates were subjected to SDS PAGE followed by western blot with 83950-5-RR (HSP90 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83950-5-PBS in a different storage buffer formulation.

  Immunohistochemical analysis of paraffin-embedded mouse testis tissue slide using 83950-5-RR (HSP90 antibody) at dilution of 1:2500 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 83950-5-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (-20°C Ethanol) fixed HepG2 cells using HSP90 antibody (83950-5-RR, Clone: 241050A10 ) at dilution of 1:250 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 83950-5-PBS in a different storage buffer formulation.

  FC experiment of Jurkat using 83950-5-RR (same clone as 83950-5-PBS)

  1x10^6 Jurkat cells were intracellularly stained with 0.25 ug HSP90 Recombinant antibody (83950-5-RR, Clone:241050A10) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 83950-5-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83950-5-RR

  Biolayer interferometry (BLl) kinetic assays of 83950-5-RR against Human HSP90 were performed. The affinity constant is 0.692 nM.

Hintergrundinformationen

HSP90, encoded by HSP90AA1, is a constitutively and ubiquitously expressed molecular chaperone that is crucial for the stability and function of many proteins. HSP90 provides chaperoning activity for client proteins; many of them are members of oncogenic pathways, indicating its implication in tumor malignancy. HSP90 mainly resides in the cytosol, while it can also be released to the extracellular space. Secreted Hsp90 is a C-terminal truncated form. It has been reported that the level of plasma Hsp90 is positively correlated with tumor malignancy in clinical cancer patients, and can be a promising diagnostic marker for tumor malignancy in clinical application. This antibody may cross-react with HSP90AB1 due to the high homology between them.
