  IHC staining of rat spleen using 30293-1-AP

  Immunohistochemical analysis of paraffin-embedded rat spleen tissue slide using 30293-1-AP (IFN Gamma antibody) at dilution of 1:200 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  FC experiment of rat splenocytes using 30293-1-AP

  1X10^6 PMA and ionomycin treated rat splenocytes cells were intracellularly stained with 0.5 ug Anti-Rat IFN Gamma (30293-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000, and 0.5 ug APC Anti-Rat CD4 (OX-35) (APC-65179, Clone: OX-35), and 0.5 ug rabbit IgG isotype control. Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Geprüfte Anwendungen

• Erfolgreiche Detektion in IHC: Rattenmilzgewebe; Hinweis: Antigendemaskierung mit TE-Puffer pH 9,0 empfohlen. (*) Wahlweise kann die Antigendemaskierung auch mit Citratpuffer pH 6,0 erfolgen.
• Erfolgreiche Detektion in FC (Intra): PMA and ionomycin treated rat splenocytes cells

IFN gamma (IFNG) is a soluble cytokine that is the only member of the type II class of IFN. It is secreted by Th1 cells, cytotoxic T cells and NK cells. The cytokine is associated with antiviral, immunoregulatory and anti-tumor properties and is a potent activator of macrophages. It plays crucial roles in pathogen clearance. Aberrant IFNG expression is associated with a number of autoinflammatory and autoimmune diseases. It has been identified in many studies as a biomarker for pleural tuberculosis (TB).
