  FC experiment of C57BL/6 Th17-polarized splenocytes using 98015-1-RR (same clone as 98015-1-PBS)

  1x10^6 untreated or PMA, Ionomycin, and Brefeldin A treated C57BL/6 Th17-polarized splenocytes were intracellularly stained with APC Anti-Mouse CD4 (GK1.5) and 0.25 ug Anti-Mouse IL-21 Rabbit Recombinant Antibody (98015-1-RR, Clone: 230267G7) and PE-Conjugated Goat Anti-Rabbit IgG(H+L).Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 98015-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 98015-1-RR

  Biolayer interferometry (BLl) kinetic assays of 98015-1-RR against Mouse IL-21 were performed. The affinity constant is 1.29 nM.

Hintergrundinformationen

Interleukin 21 (IL-21) is a cytokine produced predominantly by cluster of differentiation 4 (CD4+) T-cells and natural killer T-cells. IL-21 is preliminarily expressed in activated cluster of differentiation 4 (CD4+) T-cells and natural killer (NK) T-cells. IL-21 receptor (IL-21R) is a class I cytokine receptor, which requires dimerization with the indispensable common gamma chain (γc) subunit to bind IL-21. IL-21 plays a role in both the innate and adaptive immune responses by inducing the differentiation, proliferation and activity of multiple target cells including macrophages, natural killer cells, B cells and cytotoxic T cells. IL-21 mediates apoptosis in B-CLL cells through up-regulation of the BIM (BH3 family member) and enhances both direct and antibody-dependent cellular cytotoxicity in these cells.
