  WB analysis of mouse thymus using 17626-1-AP

  mouse thymus tissue were subjected to SDS PAGE followed by western blot with 17626-1-AP (IL-7Ra/CD127 antibody) at dilution of 1:1500 incubated at room temperature for 1.5 hours.

  WB analysis of K-562 using 17626-1-AP

  K-562 cells were subjected to SDS PAGE followed by western blot with 17626-1-AP (CD127/IL7R antibody) at dilution of 1:1500 incubated at room temperature for 1.5 hours.

  Immunofluorescent analysis of (4% PFA) fixed paraffin-embedded mouse spleen tissue using IL-7Ra/CD127 antibody (17626-1-AP) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

Hintergrundinformationen

CD127, also known as IL-7R subunit alpha (IL-7RA), is a type I membrane glycoprotein expressed on thymocytes, B cell precursors, most T cells, and some lymphoid and myeloid cells. IL-7R is a heterodimer composed of CD127 and the common-γ chain receptor (CD132). IL-7R plays critical roles in lymphocyte development and homeostasis. CD127 can also act as a receptor for thymic stromal lymphopoietin (TSLP).

Western blotting of protein from B cells of mice spleen. Protein extract was loaded on 8 % polyacrylamide gel and then transfert on nitrocellulose membrane. Nitrocellulose membrane was blocked with 5% BSA during 1h. Membrane was then incubated overnight at 4°C with 1/500 IL7Ra antibody (17626-1-AP). Finally, the membrane was incubated with anti-rabbit-HRP (clean blot IPdetection reagent from thermofisher)
