  Various lysates were subjected to SDS PAGE followed by western blot with 84922-6-RR (JUNB antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  FC experiment of HeLa using 84922-6-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug JUNB Recombinant antibody (84922-6-RR, Clone:242562G2) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

• 

  ChIP experiment of PC-12 using 84922-6-RR

  Chromatin was prepared from PC-12 cells starved overnight and treated with β-NGF (50 ng/ml) for 2 h. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of JUNB (84922-6-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR.

  Affinity and Kinetic Characterization of 84922-6-RR

  Biolayer interferometry (BLl) kinetic assays of 84922-6-RR against Human JUNB were performed. The affinity constant is 0.213 nM.

JunB is one of the components of the Activator Protein-1 (AP-1) transcription complex that have been implicated in the control ofthe G0/G1 transtion in fibroblasts. AP-1 is a collection of dimers formed by members of the Jun-, Fos-, ATF- and Maf multigene families that bind to specific DNA regulatory elements called AP-1/12-O-tetradecanoylphorbol-13-acetate-responsive elements (TREs) and cAMP-responsive elements (CREs). JunB binds to the DNA sequence 5'-TGA[CG]TCA-3', and involves in the regulation of gene activity following the primary growth factor response. It also acts either as a tumor suppressor or as an oncogene depending on the cell and physiopathological context
