  WB analysis of RAW 264.7 using 20189-1-AP

  RAW 264.7 cells were subjected to SDS PAGE followed by western blot with 20189-1-AP (MAFB Antibody) at dilution of 1:1000 incubated at room temperature for 1.5 hours.

• 

  IF Staining of U-251 using 20189-1-AP

  Immunofluorescent analysis of (4% PFA) fixed U-251 cells using MAFB antibody (20189-1-AP) at dilution of 1:1000 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-phalloidin (red).

• 

  FC experiment of RAW 264.7 using 20189-1-AP

  1X10^6 RAW 264.7 cells were intracellularly stained with 0.4 ug Anti-Human MAFB (20189-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000 (red), or 0.4 ug rabbit IgG isotype control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

Hintergrundinformationen

MAFB (MAF BZIP Transcription Factor B), also known as V-maf musculoaponeurotic fibrosarcoma oncogene homolog B, is a widely expressed basic leucine zipper transcription factor belonging to the Maf family. MAFB expression is significantly elevated in response to a broad range of metabolic and immunological stimuli that promote macrophage M2 polarization, cholesterol efflux, or both, whereas it is strongly down-regulated in response to various pro-inflammatory pathogenic triggers(PMID: 28790455). MAFB is highly expressed in tumor tissues and required for proliferation and tumorigenicity of osteosarcoma cells(PMID: 32234710) and is a marker of tumor-associated macrophages in both mouse and human(PMID: 31679694).
