  WB analysis of HEK-293 using 82958-1-RR

  WB result of MAX antibody (82958-1-RR; 1:1000; incubated at room temperature for 1.5 hours) with sh-Control and sh-MAX transfected HEK-293 cells.

  Various lysates were subjected to SDS PAGE followed by western blot with 82958-1-RR (MAX antibody) at dilution of 1:4000 incubated at room temperature for 1.5 hours.

  Chromatin was prepared from NCCIT cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of MAX (82958-1-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR.

  Biolayer interferometry (BLl) kinetic assays of 82958-1-RR against Human MAX were performed. The affinity constant is 0.257 nM.

Max, a member of MAX family, contains a basic helix-loop-helix (bHLH) domain, is a transcription regulator. MAX can form a sequence-specific DNA-binding protein complex with MYC or MAD which recognizes the core sequence 5'-CAC[GA]TG-3'. The MYC-MAX complex is a transcriptional activator, while the MAD-MAX complex is a repressor. MAX could dimerizated with another bHLH protein to form a heterodimer, such MYC or MAD. Thus bands recognized by this antibody much larger than predicted.
