  WB analysis using 83718-5-RR (same clone as 83718-5-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 83718-5-RR (MIB1 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83718-5-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 83718-5-RR (same clone as 83718-5-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug MIB1 Recombinant antibody (83718-5-RR, Clone:240676E4) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 83718-5-PBS in a different storage buffer formulation.

  1x10^6 SH-SY5Y cells were intracellularly stained with 0.25 ug MIB1 Recombinant antibody (83718-5-RR, Clone:240676E4) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue).   Affinity and Kinetic Characterization of 83718-5-RR

  Biolayer interferometry (BLl) kinetic assays of 83718-5-RR against Human MIB1 were performed. The affinity constant is 5.15 nM.

Hintergrundinformationen

MIB1(Mind bomb homolog 1) is an E3 ubiquitin-protein ligase that mediates ubiquitination of Delta receptors, which act as ligands of Notch proteins. MIB1, identified as an interactor of death associated protein kinase (DAPK), probably regulate apoptosis through ubiquitination and degradation of DAPK, thereby antagonizing anti-apoptotic effects of DAPK1 to promote TNF-induced apoptosis. MIB1 promotes proteasomal degradation of NRF2 and sensitizes lung cancer cells to ferroptosis(PMID: 34670864). In addition, MIB1 upregulates IQGAP1 and promotes pancreatic cancer progression by inducing ST7 degradation(PMID: 33793053).
