  FC experiment of HeLa using 83803-2-RR (same clone as 83803-2-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug MITF Recombinant antibody (83803-2-RR, Clone:240815E7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83803-2-PBS in a different storage buffer formulation.

  ChIP experiment of K-562 using 83803-2-RR (same clone as 83803-2-PBS)

  Chromatin was prepared from K-562 cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 20 µg of cross-linked chromatin, 5 µg of MITF (83803-2-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 83803-2-PBS in a different storage buffer formulation.

Hintergrundinformationen

The retinal pigment epithelium (RPE) has a essential role in maintaining visual function and dedifferentiation of RPE contributes to the pathophysiology of several ocular diseases (PMID:22523078). Microphthalmia-associated transcription factor (MITF) is a key regulator of RPE differentiation that is also down-regulated in dedifferentiated hfRPE cells. MITF is a basic helix-loop-helix (hHLH)-leucine zipper protein that involves in the development of various cell types, including neural crest-derived melanocytes and optic cup-derived retinal pigment epithelial cells (PMID: 10578055).
