  WB analysis using 83694-4-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 83694-4-RR (MTMR9 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours.

  IF Staining of HeLa using 83694-4-RR

  Immunofluorescent analysis of (-20°C Ethanol) fixed HeLa cells using MTMR9 antibody (83694-4-RR, Clone: 240167G7 ) at dilution of 1:200 and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) (SA00013-2).

  FC experiment of A549 using 83694-4-RR

  1x10^6 A549 cells were intracellularly stained with 0.25 ug MTMR9 Recombinant antibody (83694-4-RR, Clone:240167G7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  FC experiment of HeLa using 83694-4-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug MTMR9 Recombinant antibody (83694-4-RR, Clone:240167G7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  Affinity and Kinetic Characterization of 83694-4-RR

  Biolayer interferometry (BLl) kinetic assays of 83694-4-RR against Human MTMR9 were performed. The affinity constant is below 1 pM.

MTMR9 (Myotubularin-related protein 9) acts as an adapter for myotubularin-related phosphatases (PMID: 19038970; 22647598). MTMR9 can increase lipid phosphatase MTMR6 catalytic activity, specifically towards phosphatidylinositol 3,5-bisphosphate and MTMR6 binding affinity for phosphorylated phosphatidylinositols (PMID: 19038970; 22647598). It can also increase MTMR8 catalytic activity towards phosphatidylinositol 3-phosphate and stabilize MTMR8 protein levels (PMID: 22647598).
