  WB analysis using 84555-2-RR (same clone as 84555-2-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 84555-2-RR (OSMR antibody) at dilution of 1:1000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 84555-2-PBS in a different storage buffer formulation.

  IHC staining of mouse cerebellum using 84555-2-RR (same clone as 84555-2-PBS)

  Immunohistochemical analysis of paraffin-embedded mouse cerebellum tissue slide using 84555-2-RR (OSMR antibody) at dilution of 1:400 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 84555-2-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 84555-2-RR (same clone as 84555-2-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug OSMR Recombinant antibody (84555-2-RR, Clone:241865C8) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 84555-2-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 84555-2-RR

  Biolayer interferometry (BLl) kinetic assays of 84555-2-RR against Human OSMR were performed. The affinity constant is 0.769 nM.

Hintergrundinformationen

Oncostatin M (OSM) is a member of interleukin-6 family cytokines that modulates proliferation and differentiation in numerous cells (PMID: 22982441). OSM signals via a receptor complex comprising gp130 and the OSMR (also called OSMRβ) (PMID: 8999038). OSMR is a single-pass membrane protein that belongs to the type I cytokine receptor family. OSMR mRNA is expressed at relatively high levels in all neural cells and fibroblast, epithelial, and various tumor cell lines (PMID: 8999038). It has also been shown that OSMR associates with IL31RA to form the heterodimeric IL31 receptor (PMID: 15184896). OMSR has a calculated molecular weight of 110 kDa, with an apparent molecular weight of 150-180 kDa (PMID: 8999038; 17028186; 39266541; 34644107). A 70 kDa soluble form can be detected by western blot (PMID: 17028186).
