  WB analysis using 84970-1-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 84970-1-RR (PBK antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

• 

  IF Staining of HeLa using 84970-1-RR

  Immunofluorescent analysis of (4% PFA) fixed HeLa cells using PBK/SPK antibody (84970-1-RR, Clone: 242545A11 ) at dilution of 1:1000 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red).

• 

  FC experiment of HepG2 using 84970-1-RR

  1x10^6 HepG2 cells were intracellularly stained with 0.25 ug PBK/SPK Recombinant antibody (84970-1-RR, Clone:242545A11) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

Hintergrundinformationen

PBK(PDZ-binding kinase) is also named as TOPK, CT84, Nori-3, SPK and belongs to the MAP kinase kinase subfamily. PBK may have a role in the regulation of cellular proliferation and progression of the cell cycle. It has a characteristic cdc2ycyclin B phosphorylation site(SyT-P-X-KyR) at its N terminus,which is conserved across species, and it is phosphorylated in a cell cycle-dependent manner at mitosis, and that this phosphorylation is required for its activation(PMID:10779557).
