  WB analysis using 84162-3-RR (same clone as 84162-3-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 84162-3-RR (PDCD4 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 84162-3-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 84162-3-RR (same clone as 84162-3-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug PDCD4 Recombinant antibody (84162-3-RR, Clone:241463E3) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 84162-3-PBS in a different storage buffer formulation.

  Biolayer interferometry (BLl) kinetic assay of 84162-3-PBS against Human PDCD4 was performed. The affinity constant is 75.9 pM.

Programmed cell death 4 (Pdcd4) is a novel tumor suppressor that inhibits translation, progression and invasion. It was first identified as being differnetially upregulated during apoptosis. Pdcd4 interferes with the activity of the eukaryotic initiation factor (eIF) 4A by displacing the scaffold protein eIF4G from its binding to the RNA helicase eIF4A. The calculated molecular wight of unmodified PDCD4 protein is 52 kDa, but the modified protein is about 54-64 kDa.
