  WB analysis using 67806-1-Ig (same clone as 67806-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 67806-1-Ig (PKMYT1 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated GAPDH Monoclonal antibody (HRP-60004) as loading control. This data was developed using the same antibody clone with 67806-1-PBS in a different storage buffer formulation.

  WB analysis of MCF-7 using 67806-1-Ig (same clone as 67806-1-PBS)

  WB result of PKMYT1 antibody (67806-1-Ig; 1:2000; incubated at room temperature for 1.5 hours) with sh-Control and sh-PKMYT1 transfected MCF-7 cells. This data was developed using the same antibody clone with 67806-1-PBS in a different storage buffer formulation.

  Immunohistochemical analysis of paraffin-embedded human colon cancer tissue slide using 67806-1-Ig (PKMYT1 antibody) at dilution of 1:1000 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 67806-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

Protein kinase, membrane associated tyrosine/threonine (PKMYT1) is a member of the Wee family of protein kinases. PKMYT1 acts as a negative regulator of the cell cycle by inactivating the CDK1-cyclinB complex through phosphorylation of Tyr14/Tyr15 and preventing cell cycle entering into mitosis at the G2/M transition. In mammalian somatic cells, PKMYT1 affects the assembly of the Golgi apparatus and endoplasmic reticulum during telophase of mitosis by inhibiting cyclin activities. Abnormal expression of PKMYT1 is closely associated with multiple types of cancers. PKMYT1 protein was mainly localized in the cytoplasm. (PMID: 31695486, PMID: 32801781)
