Hintergrundinformationen
Secretory phospholipase A2 (sPLA2) are implicated in a number of crucial physiological and pathological processes in mammals. Different types of membrane receptors for sPLA2s have been identified. The M-type receptor, PLA2R1 (phospholipase A2 receptor 1, 180 kDa), is a type I transmembrane glycoprotein containing extracellularly a cysteine-rich domain, a fibronectin type II domain and eight carbohydrate-recognition domains. It is a receptor for phosholipase sPLA2-IB/PLA2G1B but not sPLA2-IIA/PLA2G2A. It is also able to bind to snake PA2-like toxins. PLA2R1 may be involved in responses in proinflammatory cytokine productions during endotoxic shock. It also has endocytic properties and rapidly internalizes sPLA2 ligands, which is particularly important for the clearance of extracellular sPLA2s to protect their potent enzymatic activities. It has been recently shown that PLA2R1 is the major target antigen in idiopathic membranous nephropathy (iMN). Patients with iMN have antibodies against a conformation-dependent epitope in PLA2R1.
