  WB analysis using 83142-4-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 83142-4-RR (PPFIA3 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  WB analysis of HEK-293 using 83142-4-RR

  WB result of PPFIA3 antibody (83142-4-RR; 1:20000; incubated at room temperature for 1.5 hours) with sh-Control and sh-PPFIA3 transfected HEK-293 cells.

  1x10^6 HEK-293 cells were intracellularly stained with 0.25 ug PPFIA3 Recombinant antibody (83142-4-RR, Clone:230459F6) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug rabbit IgG isotype control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

  FC experiment of U-251 using 83142-4-RR

  1x10^6 U-251 cells were intracellularly stained with 0.25 ug PPFIA3 Recombinant antibody (83142-4-RR, Clone:230459F6) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug rabbit IgG isotype control (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

• 

  Affinity and Kinetic Characterization of 83142-4-RR

  Biolayer interferometry (BLl) kinetic assays of 83142-4-RR against Human PPFIA3 were performed. The affinity constant is 0.131 nM.

Hintergrundinformationen

PPFIA3, also named as KIAA0654, belongs to the liprin family and Liprin-alpha subfamily. PPFIA3 may regulate the disassembly of focal adhesions. It may localize receptor-like tyrosine phosphatases type 2A at specific sites on the plasma membrane, possibly regulating their interaction with the extracellular environment and their association with substrates. PPFIA3 exhibites more than 5 fold accumulations in the monoribosome fraction. PPFIA3 is decreased at the protein level by the miR-99 family but not in the luciferase reporter with the 3'UTR of PPFIA3. (PMID:21212412)
