  Jurkat cells were subjected to SDS PAGE followed by western blot with 55143-1-AP (PSMB11 antibody) at dilution of 1:300 incubated at room temperature for 1.5 hours.

  1X10^6 MCF-7 cells were intracellularly stained with 0.2 ug Anti-Human PSMB11 (55143-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000 (red), or 0.2 ug Control Antibody. Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011).

Hintergrundinformationen

PSMB11 belongs to the peptidase T1B family. It is a multicatalytic proteinase complex which is characterized by its ability to cleave peptides with Arg, Phe, Tyr, Leu, and Glu adjacent to the leaving group at neutral or slightly basic pH. PSMB11 has an ATP-dependent proteolytic activity. Incorporated instead of PSMB5 or PSMB8, this unit reduces the chymotrypsin-like activity of the proteasome. It plays a pivotal role in development of CD8-positive T cells. The antibody is specific to PSMB11.

We tested the antibody in different dilutions on different cells of the thymus and Psmb11 knockout (KO) mice and could not obtain a specific staining with the antibody. In our hands the signal in the Psmb11-KO mice was comparable to the one obtained from either thymocytes or thymus epithelial cells of Wildtype mice .
