  WB analysis using 83024-1-RR (same clone as 83024-1-PBS)

  Jurkat cells were subjected to SDS PAGE followed by western blot with 83024-1-RR (PTPN18 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83024-1-PBS in a different storage buffer formulation.

  FC experiment of HEK-293 using 83024-1-RR (same clone as 83024-1-PBS)

  1x10^6 HEK-293 cells were intracellularly stained with 0.25 ug PTPN18 Recombinant antibody (83024-1-RR, Clone:230154G8) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83024-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83024-1-RR

  Biolayer interferometry (BLl) kinetic assays of 83024-1-RR against Human PTPN18 were performed. The affinity constant is 7.48 nM.

Hintergrundinformationen

PTPN18 (also called PTP-HSCF or BDP1), a member of the protein tyrosine phosphatase superfamily, has been reported to be a tumor suppressor in breast cancer by dephosphorylating HER2 and suppressing cell proliferation and invasion. In addition to the well-established role of PTPN18 in breast cancer, our previous study found that PTPN18 and CSK cooperate to inhibit the events triggered by SRC kinases. (PMID: 35982039)
