  WB analysis of HEK-293 using 80077-2-RR (same clone as 80077-2-PBS)

  Phosphatase inhibitor treated or λ phosphatase treated or untreated HEK-293 cells were subjected to SDS PAGE followed by western blot with 80077-2-RR (Phospho-P53 (Ser392) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with GAPDH Monoclonal antibody (60004-1-Ig) as loading control. This data was developed using the same antibody clone with 80077-2-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 80077-2-RR (same clone as 80077-2-PBS)

  1x10^6 λ phosphatase treated HeLa cells were intracellularly stained with 0.2 ug Phospho-P53 (Ser392) Recombinant Antibody (80077-2-RR, Clone:241575F6)(red) or Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9)(blue) and PE-conjugated Goat Anti-Rabbit IgG. 1x10^6 untreated HeLa cells were intracellularly stained with 0.2 ug Phospho-P53 (Ser392) Recombinant Antibody (80077-2-RR, Clone:241575F6) and PE-conjugated Goat Anti-Rabbit IgG(black). Cells were fixed with 4% PFA and permeabilized with 90% MeOH. This data was developed using the same antibody clone with 80077-2-PBS in a different storage buffer formulation.

P53 is activated in response to alteration of normal cell homeostasis, including DNA damage, nutrient starvation, heat shock, virus infection, pH change, hypoxia, and oncogene activation. P53 maintains genetic stability by regulating different processes, such as cell-cycle arrest, DNA synthesis and repair, programmed cell death, and energy metabolism. In non-stressed conditions these proteins bind p53, ubiquitylate it and target it for degradation by the proteasome. In stressed conditions the function of the MdM2-MdM4 complex is blocked by phosphorylation, protein-binding events and/or enhanced degradation. (PMID: 19935675, PMID: 24379683)
