  Various lysates were subjected to SDS PAGE followed by western blot with 26153-1-AP (SMEK1 antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours.

  IHC staining of human colon cancer using 26153-1-AP

  Immunohistochemical analysis of paraffin-embedded human colon cancer tissue slide using 26153-1-AP (SMEK1 antibody) at dilution of 1:600 (under 20x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  IF Staining of MCF-7 using 26153-1-AP

  Immunofluorescent analysis of (4% PFA) fixed MCF-7 cells using SMEK1 antibody (26153-1-AP) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-phalloidin (red).

PPP4R3A also known as SMEK1, is the regulatory subunit of serine/threonine-protein phosphatase 4. SMEK1 regulates the activity of PPP4C at centrosomal microtubule organizing centers. The PPP4C-PPP4R2-PPP4R3A PP4 complex specifically dephosphorylates H2AX phosphorylated on 'Ser-140' (gamma-H2AX) generated during DNA replication and required for DNA DSB repair(PMID: 18614045).
