  IHC staining of mouse lung using 84445-1-RR (same clone as 84445-1-PBS)

  Immunohistochemical analysis of paraffin-embedded mouse lung tissue slide using 84445-1-RR (SMURF2 antibody) at dilution of 1:1000 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 84445-1-PBS in a different storage buffer formulation.

  IF Staining of U2OS using 84445-1-RR (same clone as 84445-1-PBS)

  Immunofluorescent analysis of (4% PFA) fixed U2OS cells using SMURF2 antibody (84445-1-RR, Clone: 241486F3 ) at dilution of 1:250 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red). This data was developed using the same antibody clone with 84445-1-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed U2OS cells using SMURF2 antibody (84445-1-RR, Clone: 241486F3 ) at dilution of 1:500 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-phalloidin (red). This data was developed using the same antibody clone with 84445-1-PBS in a different storage buffer formulation.

  FC experiment of U2OS using 84445-1-RR (same clone as 84445-1-PBS)

  1x10^6 U2OS cells were intracellularly stained with 0.25 ug SMURF2 Recombinant antibody (84445-1-RR, Clone:241486F3) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 84445-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

SMURF2 is a HECT domain E3 ubiquitin ligase involved in degradation of SMADs,TGF-beta receptor,and other substrates.It also functions in regulation of neuronal and planar cell polarity, induction of senescence, and tumor suppression(PMID:22231558).SMURF2 associates constitutively with SMAD7 and it is nuclear, but binding to SMAD7 induces export and recruitment to activated TGFBR, where it causes degradation of receptors and of SMAD7 via proteasomal and lysosomal pathways(PMIF:11163210).
