  WB analysis using 68260-1-Ig (same clone as 68260-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 68260-1-Ig (SNX12 antibody) at dilution of 1:20000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated GAPDH Monoclonal antibody (HRP-60004) as loading control. This data was developed using the same antibody clone with 68260-1-PBS in a different storage buffer formulation.

  IF Staining of HeLa using 68260-1-Ig (same clone as 68260-1-PBS)

  Immunofluorescent analysis of (-20°C Ethanol) fixed HeLa cells using SNX12 antibody (68260-1-Ig, Clone: 2C6C8 ) at dilution of 1:400 and Multi-rAb CoraLite ® Plus 488-Goat Anti-Mouse Recombinant Secondary Antibody (H+L) (RGAM002), CL594-phalloidin (red). This data was developed using the same antibody clone with 68260-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

SNXs (sorting nexins) are a diverse group of cytoplasmic and membrane-associated proteins that are classified by the presence of a phospholipid-binding motif-the PX domain (PMID:12461558). SNXs are involved in endocytosis and protein trafficking. SNX12 (Sorting nexin 12) is localized in early endosomes and SNX12 overexpression specifically prevents the degradative pathway from early to late endosomes/lysosomes (PMID: 22719997). SNX12 may participate in Alzheimer's disease (AD) pathology by regulating the endocytosis of BACE1 and affecting β-processing of APP (PMID: 22709416).
