  Various lysates were subjected to SDS PAGE followed by western blot with 83787-5-RR (SON antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  1x10^6 A431 cells were intracellularly stained with 0.25 ug SON Recombinant antibody (83787-5-RR, Clone:240845B7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set.

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug SON Recombinant antibody (83787-5-RR, Clone:240845B7) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set.

  Affinity and Kinetic Characterization of 83787-5-RR

  Biolayer interferometry (BLl) kinetic assays of 83787-5-RR against Human SON were performed. The affinity constant is 0.591 nM.

SON, also called C21orf50 and DBP5, is a nuclear speckle-localized protein that shares homology with pre-mRNA splicing accessory factors (PMID: 10950926). It is an RNA-binding protein that acts as an mRNA splicing cofactor by promoting efficient splicing of transcripts that possess weak splice sites, and specifically promotes splicing of many cell-cycle and DNA-repair transcripts that possess weak splice sites, such as TUBG1, KATNB1, TUBGCP2, AURKB, PCNT, AKT1, RAD23A, and FANCG (PMID: 20581448; 21504830).
