  WB analysis of Jurkat using 66427-1-Ig (same clone as 66427-1-PBS)

  Jurkat cells were subjected to SDS PAGE followed by western blot with 66427-1-Ig (STAT5B Antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 66427-1-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed HeLa cells using STAT5B antibody (66427-1-Ig, Clone: 1D9B11 ) at dilution of 1:800 and CoraLite®488-Conjugated Goat Anti-Mouse IgG(H+L) (SA00013-1), CL594-phalloidin (red). This data was developed using the same antibody clone with 66427-1-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 66427-1-Ig (same clone as 66427-1-PBS)

  1X10^6 HeLa cells were intracellularly stained with 0.5 ug Anti-Human STAT5B (66427-1-Ig, Clone:1D9B11) (red) labeled with FlexAble CoraLite® Plus 555 Antibody Labeling Kit for Mouse IgG2a (KFA042), or 0.5 ug Mouse IgG2a Isotype Control (66360-2-Ig, Clone: K11A1B2A2) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 66427-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

STAT5B belongs to the transcription factor STAT family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. STAT5B carries out a dual function: signal transduction and activation of transcription. STAT5B mediates the signal transduction triggered by various cell ligands, such as IL2, IL4, CSF1, and different GH. It has been shown to be involved in diverse biological processes, such as TCR signaling, apoptosis, adult mammary gland development, and sexual dimorphism of liver gene expression. Despite the 95% homology between the immunogen (Ag2709) and the STAT5A protein, our WB detection showed that this antibody does not recognize the STAT5A fusion protein (Ag19546).
