  WB analysis using 68465-1-Ig (same clone as 68465-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 68465-1-Ig (STEAP4 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated GAPDH Monoclonal antibody (HRP-60004) as loading control. This data was developed using the same antibody clone with 68465-1-PBS in a different storage buffer formulation.

  FC experiment of 3T3-L1 using 68465-1-Ig (same clone as 68465-1-PBS)

  1x10^6 3T3-L1 cells were intracellularly stained with 0.25 ug STEAP4 Monoclonal antibody (68465-1-Ig, Clone:2A12G6) and CoraLite®488-Conjugated Goat Anti-Mouse IgG(H+L) (SA00013-1)(red), or 0.25 ug Mouse IgG1 isotype control Mouse McAb (66360-1-Ig, Clone: 1F8D3) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 68465-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

STEAP4 (also called STAMP2), is a member of the STEAP family. Structurally, It is a 459 amino acid protein characterized by a molecular topology of six transmembrane domains. The cytoplasmic N-terminal domain of STEAP4 shows structural similarity with bacterial and archaeal FNO oxidoreductase and STEAP4 exhibits a strong iron reductase activity. Studies in mice and human suggest that STEAP4 maybe involved in adipocyte development and metabolism, and may contribute to the normal biology of the prostate cell, as well as prostate cancer progression.
