  WB analysis using 68496-1-Ig (same clone as 68496-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 68496-1-Ig (STT3B antibody) at dilution of 1:20000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated Alpha Tubulin Monoclonal antibody (HRP-66031) as loading control. This data was developed using the same antibody clone with 68496-1-PBS in a different storage buffer formulation.

  FC experiment of A431 using 68496-1-Ig (same clone as 68496-1-PBS)

  1x10^6 A431 cells were intracellularly stained with 0.8 ug STT3B Monoclonal antibody (68496-1-Ig, Clone:3B7A8) and Multi-rAb CoraLite ® Plus 488-Goat Anti-Mouse Recombinant Secondary Antibody (H+L) (RGAM002)(red), or 0.8 ug Mouse IgG1 isotype control Mouse McAb (66360-1-Ig, Clone: 1F8D3) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 68496-1-PBS in a different storage buffer formulation.

STT3B is a component of the N-oligosaccharyl transferase (OST) enzyme which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains. N-glycosylation occurs cotranslationally and the complex associates with the Sec61 complex at the channel-forming translocon complex that mediates protein translocation across the endoplasmic reticulum (ER).
