  WB analysis using 68075-1-Ig (same clone as 68075-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 68075-1-Ig (Septin 5 antibody) at dilution of 1:20000 incubated at room temperature for 1.5 hours. The membrane was stripped and reblotted with HRP-conjugated GAPDH Monoclonal antibody (HRP-60004) as loading control. This data was developed using the same antibody clone with 68075-1-PBS in a different storage buffer formulation.

  1x10^6 Jurkat cells were intracellularly stained with 0.25 ug Septin 5 Monoclonal antibody (68075-1-Ig, Clone:1E2A3) and CoraLite®488-Conjugated Goat Anti-Mouse IgG(H+L) (SA00013-1)(red), or 0.25 ug Mouse IgG1 isotype control Mouse McAb (66360-1-Ig, Clone: 1F8D3) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C). This data was developed using the same antibody clone with 68075-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

Septin 5, a septin family member formerly called cell division control related protein 1 (CDCrel), is encoded in the 200 kb region and is abundantly expressed in the developing and adult brains of mice and humans. Although its constitutive deletion does not cause detectible structural or developmental abnormalities in the mouse brain, Septin 5 has been implicated in subtle neuronal and behavioral functions. Septin 5 is expressed in presynaptic terminals and exerts negative regulatory control over neurotransmitter release by binding to SNARE-protein, which has been implicated in neuropsychiatric disorders (PMID: 19240081). Septin 5 has 2 isoforms with the molecular mass of 39 and 43 kDa.
