  WB analysis using 67576-1-Ig (same clone as 67576-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 67576-1-Ig (TGIF2 antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 67576-1-PBS in a different storage buffer formulation.

  1x10^6 HepG2 cells were intracellularly stained with 0.4 ug TGIF2 Monoclonal antibody (67576-1-Ig, Clone:2D6B6) and CoraLite®488-Conjugated Goat Anti-Mouse IgG(H+L) (SA00013-1)(red), or 0.4 ug Mouse IgG1 isotype control Mouse McAb (66360-1-Ig, Clone: 1F8D3) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 67576-1-PBS in a different storage buffer formulation.

Hintergrundinformationen

TGIF2, is a member of a subfamily of transcription factor with a TALE(3-amino acid loop extension), an atypical homeodomain. It also contains a proline-rich region, which is a putative SH3 domain-binding site and suggested involve in transcriptional regulation. It is highly expressed in the heart, kidney, and testis, but low in the brain and prostate. Through binding directly to the 5'-CTGTCAA-3' DNA sequence or by interacting with TGF-beta activated SMAD proteins, TGIF2 acts as a transcriptional repressor. The calculated molecular weight of TGIF2 is 26 kDa, but the phosphorylated TGIF2 is about 30-35 kDa protein.
