  IHC staining of human tonsillitis using 17766-1-AP

  Immunohistochemical analysis of paraffin-embedded human tonsillitis tissue slide using 17766-1-AP (TLR3 antibody) at dilution of 1:200 (under 10x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  FC experiment of HeLa using 17766-1-AP

  1X10^6 HeLa cells were stained with 0.2ug TLR3 antibody (17766-1-AP, red) and control antibody (blue). Alexa Fluor 488-conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) with dilution 1:1500. Cells were fixed with 4% PFA and permeabilized with 0.1% Triton X-100.

Geprüfte Anwendungen

• Erfolgreiche Detektion in IHC: humanes Tonsillitisgewebe; Hinweis: Antigendemaskierung mit TE-Puffer pH 9,0 empfohlen. (*) Wahlweise kann die Antigendemaskierung auch mit Citratpuffer pH 6,0 erfolgen.
• Erfolgreiche Detektion in FC: HeLa-Zellen

• Immunhistochemie (IHC): IHC : 1:50-1:500
• Durchflusszytometrie (FC): FC : 0.20 ug per 10^6 cells in a 100 µl suspension

It is recommended that this reagent should be titrated in each testing system to obtain optimal results.

Hintergrundinformationen

TLR3 belongs to the Toll-like receptor family which are important in the innate immune response to pathogens. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities. TLR3 is a nucleotide-sensing TLR which is activated by double-stranded RNA. Upon recognition of dsRNA, TLR3 transmits signals via the adaptor protein TICAM-1/TRIF, leading to the induction of type I IFN, cytokine/chemokine production and dendritic cell (DC) maturation (PMID: 18262679).
