  WB analysis using 83704-2-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 83704-2-RR (TNPO3 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  FC experiment of HeLa using 83704-2-RR

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug TNPO3 Recombinant antibody (83704-2-RR, Clone:240728F8) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set.

  FC experiment of U-2 OS using 83704-2-RR

  1x10^6 U-2 OS were intracellularly stained with 0.25 ug TNPO3 Recombinant antibody (83704-2-RR, Clone:240728F8) and APC-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set.

  Affinity and Kinetic Characterization of 83704-2-RR

  Biolayer interferometry (BLl) kinetic assays of 83704-2-RR against Human TNPO3 were performed. The affinity constant is below 1 pM.

Transportin 3 (TNPO3) also known as IPO12, TRNSR, is a karyopherin β that works as a nuclear carrier shuttling, from the cytoplasm to the nucleus, the serine/arginine-rich proteins (PMID: 33452620). TNPO3 specifically mediates the nuclear import of splicing factor serine/arginine (SR) proteins, such as RBM4, SFRS1, and SFRS2, by recognizing phosphorylated SR domains (PMID: 11517331, 12628928). Mutations in TNPO3 are associated with Autosomal Dominant Limb-Girdle Muscular Dystrophy 2 (PMID: 23543484).
