  WB analysis using 83378-1-RR (same clone as 83378-1-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 83378-1-RR (XRN2 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 83378-1-PBS in a different storage buffer formulation.

  WB analysis of MCF-7 using 83378-1-RR (same clone as 83378-1-PBS)

  WB result of XRN2 antibody (83378-1-RR; 1:12000; incubated at room temperature for 1.5 hours) with sh-Control and sh-XRN2 transfected MCF-7 cells. This data was developed using the same antibody clone with 83378-1-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 83378-1-RR (same clone as 83378-1-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug XRN2 Recombinant antibody (83378-1-RR, Clone:240065G4) and APC-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 83378-1-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 83378-1-RR

  Biolayer interferometry (BLl) kinetic assays of 83378-1-RR against Human XRN2 were performed. The affinity constant is 1.49 nM.

Hintergrundinformationen

XRN2 is one exonuclease that degrades the Pol II associated product of poly(A) site cleavage, which is crucial for Pol II termination. During transcription termination, XRN2 cleavages at the polyadenylation site liberates a 5' fragment which is subsequently processed to form the mature mRNA and a 3' fragment which remains attached to the elongating polymerase. The processive degradation of this 3' fragment by this protein may promote termination of transcription.
