  WB analysis using 82828-14-RR (same clone as 82828-14-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 82828-14-RR (Acetyl-Histone H3 (Lys36) antibody) at dilution of 1:1000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 82828-14-PBS in a different storage buffer formulation.

  Dot Blot experiment using 82828-14-RR (same clone as 82828-14-PBS)

  Dot blot analysis was used to confirm the specificity of 82828-14-RR Acetyl-Histone H3(Lys36) antibody. peptides were spotted onto NC and probed with antibody at 1 µg/ml.The amount of peptide (μg/mL) spotted is indicated next to each row. This data was developed using the same antibody clone with 82828-14-PBS in a different storage buffer formulation.

  ChIP experiment of HeLa using 82828-14-RR (same clone as 82828-14-PBS)

  Chromatin was prepared from HeLa cells. Cells were fixed with formaldehyde for 10 minutes. The ChIP was performed with 15 µg of cross-linked chromatin, 5 µg of HIST1H3A (82828-14-RR) or 5 ug of Normal Rabbit IgG (98136-1-RR), and 20 µl of Protein A Magarose Beads. The immunoprecipitated DNA was quantified by real-time PCR. This data was developed using the same antibody clone with 82828-14-PBS in a different storage buffer formulation.

H3K36ac peaks at the 5' end of genes,manily on the two nucleosomeus immediately distal to the transcription start site, independently of gene length. H3K36ac and H3K36mac showed negative creastalk, which is mediated by GCN3 and the histone methyl transferase SDX8. Although H3K36ac is associated with gene activity, there is no linear relationship between H3K36ac and transcript levels, suggesting that H3K36ac is a binary indicator of transcription.
